Nach Besuch des Seminars Maulbronn und fünfjährigem Studium der Theologie und Philosophie in Tübingen (besonders Kant und Hegel – philosophisches Interesse blieb das ganze Leben hindurch in seiner Arbeit wirksam) und kurzer Tätigkeit als Vikar, wurde Dorner Repetent (1834), dann Professor in Tübingen (1838), Kiel (1839), Königsberg (1843, dort zugleich Mitglied des Konsistoriums und seit 1846 der Generalsynode), Bonn (1847), Göttingen (1853) und Berlin (1862). Hier wurde er auch Mitglied des Oberkirchenrats. Besondere Erwähnung verdient aus der Fülle seiner freundschaftlichen Beziehungen seine Verbindung mit dem dänischen Bischof H. L. Martensen.
Dorner war ein lebendig-gläubiger und zugleich nach wahrhaft wissenschaftlicher Begründung und Gestaltung des Glaubensinhalts strebender Theologe. Vor anderen hat er danach gerungen, auf Grund der biblischen Offenbarung unter allseitigem und umsichtigem Gebrauch der verschiedenen religiösen und wissenschaftlichen, theologischen und philosophischen Anregungen der Neuzeit, aus der Tiefe christlichen Gefühls ein ebenso harmonisches wie umfassendes Ganzes christlicher Lehrwissenschaft herzustellen, zugleich aber auch von der echt evangelischen Idee der Kirche aus an einer Neugestaltung des deutschen Kirchentums mitzuarbeiten. – Dorner sah im Pietismus die unstreitig bedeutendste Erscheinung seit der Reformation wegen seiner vertiefenden und belebenden Wirkung auf das Gemeindeleben, verteidigte aber ihm gegenüber das Recht der philosophisch-theologischen Spekulation und der Kritik auch am Inhalt der Heiligen Schrift, sofern er dem Materialprinzip des reformatorischen Rechtfertigungsglaubens nicht entspricht (Der Pietismus, insbesondere in Württemberg und seine spekulativen Gegner Binder und Merklin, 1840). – Sein eigentliches Ziel war eine deutsche evangelische Nationalkirche, die Dorner sich als einen Bund (Union) mit einer zu schaffenden gemeinsamen Bekenntnisgrundlage dachte, wobei aber die bisher geltenden Symbole nirgends anzugreifen seien. Einen Anfang dieser Entwicklung sah Dorner in dem deutschen evangelischen Kirchentag 1848 und in der Eisenacher Konferenz der Kirchenregierungen. Ihre Förderung erwartete er von einer stärkeren Beteiligung der Laienwelt an den kirchlichen Angelegenheiten auf der Ebene synodaler Einrichtungen. In diesen Zusammenhang gehört auch sein Interesse für den Gustav-Adolf-Verein, für die Innere Mission, aber auch für eine Lehrordnung, die der freieren geistigen Entwicklung innerhalb der protestantischen Kirche Spielraum gewähren sollte, endlich auch seine Ablehnung der Staatskirchenideen von F. J. Stahl und der Hengstenbergischen Reaktion (vergleiche hierzu die Sendschreiben an C. I. Nitzsch und ?Julius Müller über eine Reform der evangelischen Landeskirchen im Zusammenhang mit der Herstellung einer evangelischen deutschen Nationalkirche, 1848). – Unermüdlich widmete sich Dorner der Aufgabe der Verbesserung der lutherischen Bibelübersetzung, wobei es schon damals galt, Hemmungen des lutherischen Konfessionalismus abzuwehren (Vortrag über einheitliche Textgestaltung beziehungsweise Verbesserung der lutherischen Bibelübersetzung, gehalten auf der deutschen evangelischen Kirchenkonferenz 1868). Bemerkenswert ist in seinem „System der christlichen Glaubenslehre“ (2 Bände, 1879/81), weil neuartig und charakteristisch, die „Pisteologie“, welche die Stufen auf dem Weg zum Glauben schildert.
